Land Transport Authority (LTA) Singapore

Part of a dynamic team of transport planners who plan the future transportation infrastructure for Singapore. We are responsible for shaping the future transportation network of Singapore that includes identifying, developing and evaluating new road and rail infrastructure projects.

As part of the work, we design and supervise data collection activities, analyse and interpret survey results, develop transport network improvements proposals, forecast travel demand, assess community benefits and determine economic performance. We also work closely with other Government agencies to develop integrated transport-landuse plans and provide transportation planning advice on future land use development initiatives. Part of the role includes extensive community and grassroots engagements with stakeholders, feedback gathering and collaboration / co-creation.

Launched Gosh! Kids, a creative arts education programme (photography, art, design) and community that transforms children from passive consumers into active creators. In all we do, we want to bring our children's creativity to the next level.

I write content for Gosh! Kids and my own newsletter (Five For Your Hive), with the aim of inspiring readers to rethink their perspectives towards creativity & life.

Begun my creative journey in 2013 as a photographer (conceptual and art photography, focused on branding) and have collaborated with brands such as Apple. Many of my past work and portfolio of photographs can be found on my instagram @mathieubeth.

Project 1: Takeda - Quality Systems Specialist 

• Quality oversight for validation activities of implementation of LabWare LIMS.

• Ensure sufficient qualification efforts are in place with a focus on compliance. 

• Manage discrepancies through root cause analysis, risks management, corrective action planning and ultimately the resolution of discrepancies. 

• Ensure changes made to specifications of LabWare is managed and tracked via a quality management system (TrackWise).
   
• Collaboration with stakeholders throughout implementation lifecycle.

Project 2: Amgen - MES Engineer (Project Support)

• Manufacturing support for both Delta V and MES upgrade activities. 

• Delta V: 
- Generate MFG protocol to suit MFG needs during Delta V down time. 
- Liaise with cross function teams to ensure business and process needs are met.
- Ensure smooth transition for process prior and post down time.
- Supported upstream shop floor with guidance, usage, and reconciliation of protocols. 

• MES:
- Develop calculation spreadsheet to aid MES down time. 
- Develop validation protocol / test script for the validation of calculation spreadsheet. 
- In charge of validation efforts and ensure successful completion of validation efforts.

Involved in day to day process of mammalian cell culture fermentation (small scale) within the clean room environment while maintaining CGMP.

Involved in SOP revision and documentation for improvements. 

Involved in mentoring and providing guidance for new members on the team. Opportunity to take on the deputy shift lead position to plan out activities on the floor for the team and to ensure activities are carried out from start to end in a smooth and efficient manner.

1. Sterile inoculation work in BSC:

- Ensure the use of sterile techniques within BSC to ensure “first air” is maintained during the process carried out in the BSC.

2. Out of freeze (OOF) of mammalian cells:

- Proper thawing of cells.

- Manual cell count with use of microscope.

- Ensure proper aseptic transfer of cells into respective flask sizes followed by sampling. 

3. Inoculation expansion into flasks, single use bags (SUB) and stainless steel bioreactors.

- Inclusive of the set up of single-use bags (200L and 2000L), bag integrity testing (BIT) to ensure bags are integral before installation for usage. 

- Preparation and post-use processing of stainless steel bioreactors which includes, clean-in-place (CIP), steam-in-place (SIP), population of filters, filter integrity testing (FIT), etc. 

4. Harvesting process of cell culture.

- Setting up of depth filtration skids and flushing of filters prior to usage.

- Setting up of single-use mixer (SUM) harvest bag and bag integrity testing (BIT) to ensure bag is integral.

- Proper transfer to downstream for purification processes.

5. Media preparation.

- Carry out media preparation for inoculation related mediums. Including bottled media preparations in BSC aseptically.

- Involved in the setting up and use of single-use mixers (SUM) of different sizes to cater to the different sizes of reactors in use.

Manufacturing Biotechnologist (Upstream)

Chemical Process Technology Centre

Consists of practical operations training with a live process plant equipped with a distillation system plus control room hands on together with DCS. Exposure to 12hrs day/night shift duties where problems were simulated by the trainers and each team would have to work together to resolve the issue(s) on hand.

Over the course of this internship, proper training was given. Line tracing which is of utmost importance for the familiarity of the plant. A test was given to each student to ensure each student remembered the proper layout of the plant. Safety training like HAZOP was also inculcated into each student through the proper modules. In conclusion, the internship not only gave proper knowledge with regards to the industry, but also provides proper opportunities for each student to experience work life in a chemical plant with also proper exposures to problems that may occur within a chemical plant.
