I strongly believe that most of the infectious diseases can be prevented employing the mRNA technology that has shown tremendous impact in mitigating the Covid-19 pandemic. Similarly, this technology can be leveraged to treat several diseases including FLU, RSV and other bacterial diseases. I lead a dynamic team of four young individuals who are passionate of owing the work and delivering the portfolio projects following the company vision/roadmaps for 2022, 2023, 2024. We work relentlessly identifying the mechanism of action of vaccine candidates towards immunogenicity and reactogenicity. In parallel, we conduct high throughput immuno-assays and preclinical mice and NHP models to extensively characterize our lead vaccine candidates. Provide collaborative support to internal and external teams as subject matter expertise with a common goal of delivering the product to the market within stipulated time. Play instrumental role to have the best outcome in a multidisciplinary cross-functional collaborative team environment.

St. Jude Children's Research Hospital

St. Jude Children’s Research Hospital is one of the centers for excellence for influenza research and surveillance (CEIRS) and the World Health Organization (WHO) Collaborating Center for Influenza.
Being positioned as a lead researcher of the Infectious diseases department, I work as a part of the structural team of the Center for Influenza vaccine research for high-risk populations (CIVR-HRP). Our research is focused on the implication of HA stability in enhancing vaccine production, durability, and immunogenicity. Influenza virus hemagglutinin (HA) stability is one of the crucial factors determining viral infectivity, pathogenicity, immunogenicity, and transmissibility. We have identified several point mutations that influence the HA stability of influenza viruses followed by in vitro (pH selection) and in vivo (ferret-transmission study) selection of mutants. We employ reverse genetics to generate vaccine strains with stability mutations in HA protein designed as computationally optimized broadly reactive antigen (COBRA) in collaboration with the University of Georgia. This project will help us target the development of a universal influenza vaccine to mitigate the pathogenic potential of flu viruses and prevent a pandemic scenario.

Inflammasomes are a multi-protein cytoplasmic complexes activated upon sensing danger signals emanating both from endogenous (within cell) or exogenous (pathogenic organisms) sources. As a postdoctoral fellow at Dr. Kanneganti lab I am involved in studying and understanding the innate molecular underpinnings of chronic autoinflammatory diseases using mouse models and also to evaluate the role of novel NOD-like receptors in the context of infectious diseases, and colorectal carcinogenesis.

As a postdoctoral research scientist, I lead the study to understand the role of NLRP3 inflammasome activation in the pathophysiology of cardiovascular disease and cryopyrinopathies. We unveiled the role of mitochondria as a scaffolding platform required for NLRP3 inflammasome activation. Specifically, mitochondrial cardiolipin present in the inner leaflet gets externalized during priming events and recruits caspase-1 and NLRP3 molecules onto mitochondria. This step is followed by activation signals which mobilize ca+2 signaling to drive the adapter molecule ASC to bind to NLRP3 and Caspase-1 to form an active inflammasome complex. I have followed up on this research to further investigate the role of these adapter molecules in driving inflammasome assembly and activation during cryo-pyrinopathies. We have employed gain-of-function NLRP3 mouse models to study these effects. In collorary projects, I have worked on NLRP12 molecule contribution to elicit innate immune responses in response to Influenza viruses. In particular, we found that NLRP12 potentiates the recruitment of immune cells to the site of infection via the expression of chemokine KC/CXCL1.
