In hopes of finding efficient novel drug targets to treat aquaporin (AQP)-related diseases including brain oedema, I analysed and investigated the effect of phosphorylation on the trafficking (movement) of mammalian AQPs between the cytoplasm and plasma membrane. The hypotheses, research question, aims and objectives of the project were successfully met through a series of intensive laboratory experiments as well as bioinformatic and statistical analysis. The results, some of which were ground-breaking and reported for the first time, were presented in the form of a written dissertation as well as an in-person progress presentation in front of both the project supervisor and internal assessor.

Key lab techniques included: Agarose gel electrophoresis, DNA miniprep, DNA sequencing (Sanger), PCR, site-directed mutagenesis (QuikChange & Phusion), adherent mammalian cell culture (using HEK293 cells), restriction enzyme digests (single & double), E. coli transformation, transient transfection of mammalian AQP2 and AQP4 tagged with a green fluorescent protein (GFP) into HEK293 cells, HEK293 cell passaging, fluorescence microscopy (to check confluence and 'health state' of cultured HEK293 cells) & confocal microscopy (to analyse AQP trafficking and carry out water experiments before (isotonic) and after (hypotonic) addition of water).
