Project title: CAPACITOR (Cardio-Pulmonary Polysaccharide based nanoparticles as theranostic tools in heart failure)

Summary: Heart failure with preserved ejection fraction (HFpEF) is a complex syndrome that disproportionately affects women
and the elderly, and accounts for half of all heart failure cases. HFpEF long-term prognosis remains poor and mortality rates are unacceptably high. Clinical trials employing conventional heart failure therapy in HFpEF patients have yielded unsatisfactory results. The lack of effective treatments for HFpEF is a primary clinical concern in cardiology that must be addressed urgently. Despite the effectiveness of previously identified potential treatments for HFpEF such as miRNAs, BNP, and proANP31-67, the use of these compounds is currently limited because of their lower bioavailability. To overcome this drawback, we conceived CAPACITOR, the idea of using a Polysaccharide-based nanotechnology delivery
system to enhance the efficacy of drugs and to target the heart.

Project title: Non-coding RNA in treatment and diagnosis of heart failure with preserved ejection fraction (HFpEF)

Summary: Heart failure with preserved ejection fraction (HFpEF) is a complex syndrome that disproportionately affects women and the, and accounts for half of all heart failure cases. HFpEF long-term prognosis remains poor and mortality rates are unacceptably high. Clinical trials employing classical heart failure therapy in HFpEF patients have shown disappointing results and the lack of treatments for HFpEF represents a growing unmet need in cardiology. One critical obstacle to therapeutic innovation in HFpEF has been the absence of a comprehensive characterization of pre-clinical models. Thus, we have conducted a detailed investigation of clinically relevant HFpEF models, including high-throughput cardiac and blood screening and multi-organ phenotyping, to better understand the regulatory mechanisms underlying the pathophysiology of this complex syndrome. We, then, developed a dedicated framework for data integration using cutting-edge analytical methods and identified novel circulating and cardiac microRNA signatures for HFpEF.

Project title: Inhalable Calcium Phosphate nanoparticles as siRNA carrier: towards anti-inflammatory nanomedicine in cardiovascular diseases
Summary: In general, during this thesis, we provided a wide range of chemical to functional studies. Our proof of concept studies revealed that CaP nanoparticles, obtained from the developed protocol in the CUPIDO project, are well-tolerated, non-toxic, and biocompatible. Utilization of CaP nanoparticles provides an efficient delivery system for siRNA modality. We also showed the high potential of OSM-specific siRNA-loaded CaP nanoparticles in silencing OSM gene for the potential resolution of inflammation. In this proof of concept study, these results pave the way for innovative use of CaP nanoparticles as a theranostic tool.
