When I was at the bench, I was lucky to live through two major technological advancements. 1. Almost overnight, next-generation DNA sequencing technology changed our ability to read DNA. "Don't think, just sequence" became our new lab phrase. 2. I also worked with several different genome editing tools. First, zinc finger nucleases, then TALENs, and then (BOOM) CRISPR. Suddenly, CRISPR had made genome editing easy. --- But one thing stayed the same during that time: DNA synthesis. The more high-throughput our ability to sequence and edit DNA became, the more problems we noticed with the synthetic DNA we were buying. And that's because that DNA synthesis tech was stuck. ➟ We couldn't order many synthetic genes. ➟ The DNA we ordered was very error-prone. ➟ Phosphoramidite synthesis generates vast amounts of solvent waste. --- So, as frustrated scientists, we set out to develop a new DNA synthesis technology and a great place to work.
