Involved in day to day process of mammalian cell culture fermentation (small scale) within the clean room environment while maintaining CGMP.

Involved in SOP revision and documentation for improvements. 

Involved in mentoring and providing guidance for new members on the team. Opportunity to take on the deputy shift lead position to plan out activities on the floor for the team and to ensure activities are carried out from start to end in a smooth and efficient manner.

1. Sterile inoculation work in BSC:

- Ensure the use of sterile techniques within BSC to ensure “first air” is maintained during the process carried out in the BSC.

2. Out of freeze (OOF) of mammalian cells:

- Proper thawing of cells.

- Manual cell count with use of microscope.

- Ensure proper aseptic transfer of cells into respective flask sizes followed by sampling. 

3. Inoculation expansion into flasks, single use bags (SUB) and stainless steel bioreactors.

- Inclusive of the set up of single-use bags (200L and 2000L), bag integrity testing (BIT) to ensure bags are integral before installation for usage. 

- Preparation and post-use processing of stainless steel bioreactors which includes, clean-in-place (CIP), steam-in-place (SIP), population of filters, filter integrity testing (FIT), etc. 

4. Harvesting process of cell culture.

- Setting up of depth filtration skids and flushing of filters prior to usage.

- Setting up of single-use mixer (SUM) harvest bag and bag integrity testing (BIT) to ensure bag is integral.

- Proper transfer to downstream for purification processes.

5. Media preparation.

- Carry out media preparation for inoculation related mediums. Including bottled media preparations in BSC aseptically.

- Involved in the setting up and use of single-use mixers (SUM) of different sizes to cater to the different sizes of reactors in use.

Consists of practical operations training with a live process plant equipped with a distillation system plus control room hands on together with DCS. Exposure to 12hrs day/night shift duties where problems were simulated by the trainers and each team would have to work together to resolve the issue(s) on hand.

Over the course of this internship, proper training was given. Line tracing which is of utmost importance for the familiarity of the plant. A test was given to each student to ensure each student remembered the proper layout of the plant. Safety training like HAZOP was also inculcated into each student through the proper modules. In conclusion, the internship not only gave proper knowledge with regards to the industry, but also provides proper opportunities for each student to experience work life in a chemical plant with also proper exposures to problems that may occur within a chemical plant.
